Mechanism of Pressor Action of Metoclopramide






Abstract
The pressor response of metoclopramide in the rat was investigated in the present study.
Meto.lopramide administered into a femoral vein of the rat produced dose-related pressor ac-don secondarily followed by depressor action (unpublished result).
This hypertensive response evoked by metoclopramide was inhibited by pretreatment with atropine, chlorisondamine and prazosin, while was not influenced by cyproheptadine or clonidine. Metoclopramide also did not affect the pressor response of dopamine.
Theses experimental results suggest that metoclopramide induces the pressor response through the nicotinic and muscarinic action in autonomic ganglia without affecting dopaminergic receptors.
